Step-by-Step Operation (Izon AFC V2)
Preparation: Place the carousel (2 mL or 5 mL) on the AFC and ensure the nozzle is aligned with the column.
Calibration: Run the scale calibration (using the 10g weight) and verify the carousel alignment.
Set Parameters: Use the touch screen to select the column type and set the desired buffer volume and collection volumes.
Load Column & Sample: Mount the column (remove the top cap first to prevent damage). Load the sample, ensuring no residual buffer is left above the frit.
Start Run: Press start. The device will first collect the buffer volume (red light) and then move to fraction collection.
Collection: The carousel will automatically rotate to collect fractions, and the device will alert you if you need to load a second carousel for more samples.
Cleanup: After the run, clean the column and flush the nozzle with DI water to prevent salt buildup.
